AAV capsid DNA engineering for Insect cell production 

We engineer capsid DNA for production of AAV in an insect cell system. Each engineered capsid construct will be fully analyzed (silver staining, CE-SDS, EM, e.g.) and, if available, compared to mammalian cell produced capsid. We will particularly focus on the composition of capsid proteins produce in insect cells compare to  mammalian cells.

Processing development for large scale AAV Manufacturing 

For each new project we will optimize the large scale production to optimize both upstream and downstream processes in order to achieve the maximum yield and full/empty ratio. SOP will develop to facilitate the transfer for GMP production. 

Medium scale AAV manufacturing: 10L XDR10 or 25L wave bioreactor 

Our medium scale production can be manufactured in 2 different vessels; 10L XDR10 bioreactor or 25L Wave. The 10L bioreactors are used for pre-200L run and to test the upstream and downstream processes as well as to produce preliminary sample product for the client.
The Wave bioreactors are used solely for final medium scale production.  

Large scale AAV manufacturing: 2x 200L XDR200 bioreactors

For our large scale AAV production, we use 200L XDR200 bioreactors and AKTA pilot for upstream and downstream process. The production strategy is developed in order to be easily transferable to a GMP facility. We use pre-designed, gamma-irradiated tubing sets as part of a closed disposable system.

Analytical and QC lab on site

The analytical lab determines the best engineered capsid for insect cell production and identifies the best method for the large scale production.
The QC lab uses physical and biological assays to ensure that the quality of the final product passes our strict internal standards and to control level of impurities and contaminants.

To initiate a new project follow these steps.

Step 1 -  Email Anna.Ucher@umassmed.edu  a brief description of your project request.  If you are a student or PostDoc, please indicate the PI's laboratory to which you belong.

Step 2 - The Director or one of the laboratory staff will contact you to schedule a consultation and to confirm approval of your project.

Step 3 - Consultation will be via Zoom, email or by phone.

Step 4 - Upon project acceptance and assignment to a staff member, future consultation and sample submission for your project will continue via the assigned staff member.

Step 5 - New projects must be initiated by following Step 1.
