Fee for service contracts

If you are in need of expertise for generating and tracking defined and complex variant libraries using sequencing approaches in any genetic context and/or selection for binding properties using yeast display, we provide fee for service work where your data and your results are strictly for your use. Contracts (including confidentiality agreements) between UMass Chan and interested individuals and organizations are worked out for fee for service work.

Antibody engineering
We have extensive expertise in advanced antibody engineering approaches utilizing yeast display of Fab’s combined with custom mutagenesis (e.g. saturation mutagenesis at positions in CDR’s), flow cytometry, and next-generation sequencing readouts. Expertise for these projects is led by Prof. Daniel Bolon who pioneered the first systematic deep mutational scanning method. Services available include affinity maturation and paratope mapping utilizing rapid and cost effective barcoded sequencing readouts.

We generate and track the variants that you want
We have an extensive track record of getting mutagenesis projects with deep sequencing readouts done right the first time. We have proven strategies to rapidly and efficiently generate custom libraries of mutations in CDR’s including all single and/or double and/or triple mutations. We routinely sample more than 100 million variants of Fab’s in yeast display with rapid selection using flow cytometry of the strongest binders under multiple conditions. We have established experimental and bioinformatic pipelines for accurate identification of up to 100,000 selected variants and effective estimates of their frequency.
