Fee-for-Service Research Partnerships
We provide contract-based, fee-for-service research for investigators and organizations seeking expertise in:
- Design and generation of defined or highly complex variant libraries
- Deep sequencing–based tracking of variants in any genetic context
- Yeast display–based selection for binding properties
All projects are conducted under formal agreements between UMass Chan and the sponsoring organization, including confidentiality agreements as needed. Your data and results remain strictly yours.
Antibody Engineering Services
Our group offers advanced antibody engineering capabilities centered on yeast display of Fabs combined with custom mutagenesis, high-resolution flow cytometry, and next-generation sequencing readouts.
Scientific leadership is provided by Prof. Daniel Bolon, who pioneered the first systematic deep mutational scanning method.
Core Capabilities
- Affinity maturation
- Paratope mapping by systematic mutational scanning of CDRs
- Epitope mapping
- Barcoded deep sequencing readouts for rapid, cost-effective analysis
We specialize in designing and executing mutagenesis strategies that work the first time. Our team has extensive experience generating precisely defined mutation libraries, including:
- Complete single-mutant libraries
- Comprehensive double- and triple-mutant CDR libraries
- Saturation mutagenesis at targeted CDR positions
We routinely construct and screen libraries exceeding 100 million Fab variants using yeast display. Selection of high-affinity binders is performed under multiple conditions using flow cytometry, enabling robust and reproducible enrichment.
Our integrated experimental and bioinformatic pipelines support:
- Accurate identification of up to 100,000 selected variants
- Reliable quantitative frequency estimation
- High-confidence interpretation of mutational effects
