Development of Microscopy Techniques for Biomedical Imaging                                                                                Clive's Picture

My work involves the design of components to improve and extend the performance of the epi-fluorescence, total internal reflection (TIRF), and Biorad two photon microscopes in the Biomedical Imaging Group to answer the biological problems of interest to our collaborators.

TIRF Microscope

The microscope has been extensively use this year for the following programs:

Silvia Corvera and Debrah Leonard, UMass, to observe the specialization of plasma membrane domains for clathrin mediated endocytosis of transferrin.

Shaohui Huang, UMass, to observe how insulin regulates GluT4 vesicular trafficking near the plasma membrane of 3T3-L1 adipocytes.

John Walsh and Valerie DeCrescenzo, UMass, to observe exocytosis in nerve terminals and chromaffin cells.

Zheng Zheng Bao to image the action potential propagating across chicken embryo hearts in a bath containing a voltage sensitive fluorescent dye.

Xueyi Li, Mass General, to image GFP labeled Huntingtonâs protein.

Biorad Two Photon Microscope

A Biorad 2100MP two photon system has been installed and aligned. A new microscope stage has been constructed and a study of dendritic cells and mouse lymph nodes is under way in collaboration with Julie Mangada and Agata Jurczyk of UMass. A heated stage has been developed for the live mouse and a clamping system for the mouse skin flap is being made.
