Rotation Projects

1.   Investigate the signaling pathways important for IRS-1 and IRS-2 function.  Site-directed mutagenesis will be used to introduce specific mutations into the IRS-1 and IRS-2 genes.  Mutated genes will be expressed in mammary tumor cell lines and functional assays to assess invasion and survival will be performed.

2.   Investigate the involvement of the IRS proteins in HER2/ErbB2 signaling and resistance to therapy.  The expression and function of the IRS proteins will be evaluated in cell lines that overexpress HER2/ErbB2.  The impact on Herceptin resistance will also be assessed.

3.   Investigate crosstalk between the alpha6/beta4 integrin and IGF-1R signaling.  The effect of cooperative signaling on breast carcinoma cell function will be evaluated.

4.   Evaluate the importance of IRS-1 and IRS-2 regulated genes for mammary tumor progression.  We have identified several genes that are up- or downregulated in response to IRS-1 or IRS-2 signaling.  The expression of these genes will be validated and their functional contribution to breast carcinoma invasion and survival will be examined
