For our studies, it is useful to have a convenient way to replace bacterial genes with genes from other organisms encoding similar functions, or with mutant alleles. Simple replacements, involving no additional genes (i.e. selectable markers), are particularly valuable. Existing methods for making such replacements are effective, but cumbersome. In this project, the student would develop a set of reagents (plasmids, primers) which would make gene replacements in two steps: first, replacing the gene in question with gfp (green fluorescent protein), identifying recombinants by their green color; second, replacing gfp with the mutant or variant of choice, identifying recombinants by their loss of green color.
