Rotation Projects:

  1. We have a project to investigate the biophysical properties of an ALS associated protein.  Mutations in this protein cause it to mislocalize and aggregate in a cell culture model.  The next steps of this project entail in vitro expression and purification of the mutant proteins, and subsequent characterization of their conformational stabilities and aggregation propensities.
  2. We recently identified a novel protein interaction that is specific for mutant SOD1 proteins relative to the wild-type protein.  This interacting protein is phosphorylated in cells.  The next steps of this project entail purifying the phosphorylated protein, comparing the affinity of the phosphorylated protein versus the non-phosphorylated protein for mutant SOD1 proteins, and preparing purified phosphorylated protein for NMR and X-ray crystallographic studies.
